<trans-abstract xml:lang="en"><p>To obtain catalytically active recombinant biotin ligase (rBirA), we developed an approach for heterologous expression of BirA-MBP-fusion protein in E. coli cells, its purification and subsequent rBirA release by highly specific proteolysis using TEV protease. The recombinant protein is obtained in homogeneous state and retains specific catalytic activity. Enzyme immobilization on magnetic microparticles has allowed to develop an approach for effective enzymatic biotinylation combined with purification of target protein molecules on the affinity matrix.</p></trans-abstract><kwd-group xml:lang="ru"><kwd>BirA биотин лигаза</kwd><kwd>белок MBP</kwd><kwd>молекулярное клонирование</kwd><kwd>гетерологическая экспрессия</kwd><kwd>сайт-специфичное биотинилирование</kwd></kwd-group><kwd-group xml:lang="en"><kwd>BirA biotin ligase</kwd><kwd>MBP protein</kwd><kwd>molecular cloning</kwd><kwd>heterologous expression</kwd><kwd>site-specific biotinylation</kwd></kwd-group><funding-group><funding-statement xml:lang="ru">Работа выполнена при частичной поддержке Белорусского ремпубликанского фонда фундаментальных исследований (грант Х16Р-062 и Х17М-012).</funding-statement><funding-statement xml:lang="en">This work was partially supported by the Belarussian Foundation for Fundamental Research (grant Х16Р-062 and Х17М-012).</funding-statement></funding-group></article-meta></front><back><ref-list><title>References</title><ref id="cit1"><label>1</label><citation-alternatives><mixed-citation xml:lang="ru">Selective labeling of proteins with chemical probes in living cells / M.
