<trans-abstract xml:lang="en"><p>The Sso7d protein has exceptional structural stability and the ability to bind highly specifically to DNA, which makes the protein a promising module for creating fusion proteins and test systems. Sso7d is a part of fusion high-fidelity DNA polymerases capable of carrying out the polymerase chain reaction even in the presence of PCR inhibitors. Application of faster, simpler, and more efficient method for protein production will significantly reduce the cost of creating biosensors and conducting analyzes. This paper describes a new efficient method for obtaining recombinant Sso7d protein with a high degree of purity without using affinity chromatography.</p></trans-abstract><kwd-group xml:lang="ru"><kwd>Sso7d</kwd><kwd>ДНК-связывающий белок</kwd><kwd>гетерологическая экспрессия</kwd><kwd>выделение белка</kwd><kwd>ультрафильтрация</kwd><kwd>рекомбинантный белок</kwd></kwd-group><kwd-group xml:lang="en"><kwd>Sso7d</kwd><kwd>heterologous expression</kwd><kwd>DNA-binding proteins</kwd><kwd>protein isolation</kwd><kwd>ultrafiltration</kwd><kwd>recombinant protein</kwd></kwd-group><funding-group><funding-statement xml:lang="ru">Исследование выполнено при финансовой поддержке Национальной академии наук Беларуси (грант № 2022-27-009).</funding-statement><funding-statement xml:lang="en">The study was financially supported by the National Academy of Sciences of Belarus (grant no. 2022-27-009).</funding-statement></funding-group></article-meta></front><back><ref-list><title>References</title><ref id="cit1"><label>1</label><citation-alternatives><mixed-citation xml:lang="ru">Solution structure and DNA-binding properties of a thermostable protein from the archaeon Sulfolobus solfataricus / H.
