МОЛЕКУЛЯРНОЕ КЛОНИРОВАНИЕ, ГЕТЕРОЛОГИЧЕСКАЯ ЭКСПРЕССИЯ И ПОЛУЧЕНИЕ РЕКОМБИНАНТНОГО СОМАТОТРОПИНА GALLUS GALLUS

Соматотропины относятся к группе полипептидных гормонов, запускающих различные каскады биохимиче­ских процессов. Основная функция соматотропина – влияние на анаболические и катаболические процессы орга­низма, а также иммуномодулирующая функция. В представленной работе проведено молекулярное клонирование, гетерологическая экспрессия и очистка рекомбинантного соматотропина Gallus gallus. Созданы две технологиче­ские схемы получения соматотропина: бактериальная экспрессия с последующей солюбилизацией телец включения и периплазматическая экспрессия соматотропина в бактериальных клетках. Выход целевого гормона составил 2,43 и 15 мг белка на литр культуральной среды соответственно. В работе показано, что наиболее оптимальной является стратегия периплазматической экспрессии, при которой нет необходимости в сложных процедурах солюбилизации и рефолдинга и получаемый белковый продукт характеризуется более высоким выходом и степенью очистки.
